SORTING PROCESS - WHAT TO EXPECT

DURING YOUR SORT

  • Sort Logic 
    • Before a sort will be started, the sort logic will be reviewed (gating scheme, plot setup, intended populations to sort, etc.) Please make sure you are available for this review. Failure to review the sort logic will result in the sort being canceled and the user billed for the scheduled time of the sort.
  • Contact Information
    • After the sort is started, provide the operator with contact information so you can be contacted if something arises during the sort or for notification of completion of the sort.

AFTER YOUR SORT

  • Post Sort Purity Check
    • Once a sample is sorted, investigators can request to have a Post Sort performed. A Post Sort is a check of the purity of your collected population(s) using the gating scheme with which the sort was conducted. Each collection tube is run on the sorter and a default 2,000 events are recorded (more or less can be recorded on request.) The machine is cleaned before the post sort and between each sample run for the post sort.
    • The FCR recommends conducting a post-sort analysis whenever feasible. However, certain experimental constraints may preclude this step. These limitations include limited cell recovery, sorting directly into lysis buffer, or sorting into multi-well plates (although alternative methodologies can be explored for plate-based sorting if desired).
